I personally don’t think anime is the saddest thing that exists because there are a lot of other things that are sadder then anime. For me, anime is just another type of entertainment that japan offers. It is no different from, for example, Nickelodeon cartoons, Cartoon Network cartoons, reality T.V. shows etc.

The Attack on Titan universe is a cruel one, and Levi Ackerman might've been the character who suffered the most. Levi Ackerman has a lot going for him. He is by far one of the most popular characters in Attack on Titan, proven by the fact that he's topped two consecutive popularity polls by a landslide.
Isayama-san once said that Levi is extremely emotional and depressed and once even thought about killing himself, only to get back up and fight for humanity.
Whether it's the manga or the anime, there is no shortage of sad moments from Attack on Titan. Here are 10 of the saddest moments in the series. There is perhaps no anime series that is more gut-wrenching and intense than Attack on Titan.
However, to answer the question, I would say that Levi had the saddest backstory.
Not only did this lead to his brother becoming a villain, but his mother also had a mental breakdown and harmed him, which left him with a permanent burn scar because of how much he looks like his father. As such, when it comes to My Hero Academia's tragic backstories, Shoto seems to have suffered the most.
2 Eri. Eri's backstory is very much the perfect storm of the tragic elements commonly seen in a My Hero Academia character's backstory. She has dead parents, an abusive childhood, and had a traumatic awakening of her Quirk.
Once Katsuki developed his Quirk, his aggressive tendencies transformed into heinous bullying. He was constantly praised for his powerful Quirk and other talents. As a result of this, he developed a twisted superiority complex as a young child.
His father then orders him to be abandoned. Hyakkimaru is later saved and raised by the doctor Jukai, who gives him artificial prosthetics to help him thrive. Forced to suffer for the sake of another man’s ambition, Hyakkimaru is cursed with a fate worse than death.
11 Kyo Sohma – Fruits Basket. Being born into the Sohma family, one that has its roots in certain people being born as one of the zodiac animals, is already hard enough in Fruits Basket . However, what makes this worse is that Kyo happens to be the cat, which is the most hated zodiac animal.
When it comes to the backstories of One Piece characters, Nico Robin’s is by far one of the most tragic. When she was two years old, her mother left Ohara leaving her vulnerable to both verbal and physical abuse from her aunt, as well as other children.
Akame may appear as a cold-blooded assassin to those who do not know her, but the truth behind her character runs deeper. After her parents sold both her and her younger sister, Kurome, to the empire, she was forced to survive, even if it meant killing others.
12 Killua Zoldyck – Hunter X Hunter. Killua was born into a family of assassins, which means his childhood was anything but ordinary. At the age of three, his parents put him in dangerous situations ranging from various forms of torture to seeing if he can dodge lethal attacks.
It turns out that Ray knew everything all along, and while plotting his escape from Gracefield House since he was a child, he knew Isabella, his own mother, was feeding the children to demons.
14 Ray – The Promised Neverland. When fans first see Ray, they think of him as one of the calmest children amongst Emma and Norman. However, Ray is perhaps one of the most tragic characters in the bunch, despite The Promised Neverland being full of tragic characters.
Even if you do spit it back out. But when you experience sadness in anime? this is what helps you relate or empathize with certain characters . Sadness makes the dullest characters some of the brightest and most interesting.
17. Ai Enma (Hell Girl) Ai Enma is the “hell girl” who shows up after 12AM if called upon, and sends sinners straight to hell. The anime itself is dark and portrays the darkest parts of what humans are capable of. But Ai Enma in particular has a past few would be able to survive.
2. Revy Rebecca (Black Lagoon) Rape, abuse, murder…. These 3 words paint a dark, bloody picture of Revy’s past life. Revy is cold-hearted and at times, seems like she no longer even has a soul. But I guess that’s what life does to you when you’re forced into the dark abyss of the underworld.
Anzu (Hinamatsuri) Anzu from Hinamatsuri is a special case. The anime uses her character to portray homelessness, and the seriousness of how it affects people. When Anzu is homeless, she steals food, drinks, money, whatever it takes to survive. She’s just doing what she has to do, and her intentions are pure.
Asahi Kobe (Happy Sugar Life) Asahi is probably the most tragic character in the entire series because of how it starts and ends (and his past). He and his sister grow up in an abusive household. Him being the older brother, the abusive father constantly abuses him, the mum, and makes home life a living hell.
Jellal is a child slave, raised in the tower of heaven. Forced to do hard labor for nothing in return. Until the fateful day when Jellal and other childhood slaves forge a plan to escape. 9. Erza Scarlet ( Fairy Tail) Like Jellal, Erza also grew up a childhood slave, and was one of Jellal ’s many friends.
Making her suffering beyond what we see with most anime characters. 18. Miwa ( Wonder Egg Priority) Miwa is only a side character, if you can call her that.
These Are The Most Miserable Anime Characters (SPOILERS): 1. Homura Akemi ( Madoka Magica) Homura Akemi is either indifferent in her attitude, or plain miserable and depressing. She’s aloof and has a dark “aura” that’s hard to ignore. And that’s because of her depressing, painful back story to how she becomes a magical girl.
Ryuko Matoi ( Kill La Kill) Ryuko Matoi has every reason to be angry, miserable and pissed off. She’s on the hunt for her fathers killer, after all. It becomes her #1 mission in life. And nothing else matters in the plot of Kill La Kill. Especially during the beginning phase of the series.
Otori Asuka (Magical Girl: Spec Ops Asuka) Otori Asuka, one of the newest anime girls in the industry, represents the “ugly side” of being a magical girl. Spec Ops Asuka is a Seinen magical girl series. So it depicts the “aftermath” of saving the world as a magical girl.
Medusa made Crona through her own disturbed experiments, and doesn’t care whether Crona lives or dies during the plot in the anime series. She’s a tool to be used for Medusa’s personal gain. And nothing more. That makes Crona devoid of love, affection and warmth.
2. Crona (Soul Eater) Crona is another character who’s indifferent, angry, or aloof and depressed most of the time. In Crona ’s case, this is the result of being neglected as a child by Medusa (The Witch).
That’s where Asuka’s unhappiness stems from. Asuka has some form of PTSD because of all the stress she’s gone through as a teenager. And all the trauma associated with death, murder, destruction and the pain of losing people she loves. You can empathize with Asuka based on that by itself.
Somewhere along the way she meets a woman named Teresa, who’s a claymore (half human, half demon). After following her around and becoming attached to Teresa, the worst thing happens. And Teresa is beheaded by another Claymore. Then Clare becomes a Claymore herself by combining Teresa ’s DNA with her own (basically).
I've always thought that Kakashi's backstory is one of the saddest because he knew his dad, but then he committed suicide. It's worse to know somebody and for them to die, then not knowing them at all because you know there love, then you lose it. Obito "died" leaving him with the responsibility of Rin.

Easily the most powerful of Spike Spiegel's rogue's gallery, Mad Pierrot was also the most deranged. It is with good reason: He was captured and experimented on by a number of intelligent military scientists, breaking his mind and traumatizing it beyond repair.
Meruem was the former leader of the NGL before being consumed by the chimera ants and reborn from their queen. Consequently, he knew only violence as the freshly coronated king went about destroying the land he had once called home and murdering its denizens savagely.
Obito's backstory is more painful than Naruto's on account of the way events had transpired. Instead of being an outcast who gradually became a hero, the converse was true for him, watching helplessly as his identity was stripped away and remade through the careful machinations of Zetsu and Madara.
While Rui may have been at least partially responsible for the deal he struck to regain his legs, he was a child when he made the decision. This justifies his lapse in judgment and forces him to live with the fact that he had turned against his biological family— thus compelling him to create a fake new one from the victims he had converted.

The Dio of JoJo 's Phantom Blood arc may have been a sadistic monster, but his origins at least made him a partially sympathetic character (despite Speedwagon's claims to the contrary).
While anime villains may be notorious for being morally evil and possessing no redeeming qualities, this is not always the case. Sometimes, antagonists are presented in a way that renders them sympathetic evils, explaining their backstory well and even giving the hero pause as to whether or not they should proceed with killing them.
