Not quite certain if it was the very first but he used some sort of Conquerer's Haki against Motobaro (Duval's bull) in Anime episode 388. There may have been other subtle instances before that wasn't noticed though. Like when Luffy and Zoro sensed more than one person in Blackbeard in Mock town, implying some sort of observation Haki.
